    陈祥仁[1] 黄霄红[2] 徐宝多[3] 卢英华[3][1]厦门大学生命科学学院,福建厦门361005 [2]厦门大学医学院药学系,福建厦门361005 [3]厦门大学化学化工学院化学工程与生物工程系,福建厦门361005摘 要:利用SIH合成培养基,在7L发酵罐培养盘基网柄菌.培养147h后,细胞密度达到4.0×10^7mL^-1,为在复杂培养基上所能达到的细胞密度的2~4倍.培养过程中葡萄糖的消耗量为6.7g/L,产氨浓度达0.88g/L.对培养基中的氨基酸分析表明,赖氨酸、色氨酸、甲硫氨酸和苯丙氨酸消耗较快,显示SIH培养基的氨基酸成分还可进一步优化.采用基于Monod生长动力学的半经验模型可很好模拟细胞生长和底物消耗,并估计出动力学参数μmax=0.115h^-1,Nmax=6.0×10^7mL^-1.本研究为进一步优化合成培养基和为利用这一新型真核表达系统大规模生产重组异源蛋白奠定了基础.[著者文摘]

    High Cell Density Cultivation of Dictyostelium in Conventional BioreactorCHEN Xiang-ren, HUANG Xiao-hong, XU Bao-duo, LU Ying-hua 1. School of Life Science, Xiamen Univ. , 2. Department of Chemical and Biochemical Engineering, College of Chemistry and Chemical Engineering, Xiamen Univ. , Xiamen 361005, ChinaAbstract:Dictyostelium discoideum was cultivated on a novel synthetic SIH medium in 7 L bioreactor. After 147 h cultivation, cell density was reached up to 4.0×10^7 mL^-1. It was about 2~4 times higher than that could be obtained in axenic complex medium. Up to 6.7 g/L glucose was consumed and 0.88 g/L ammonium was produced during the cultivation, respectively. Analysis of samples with respect to amino acids indicated that lysine, tryptophan, methionine and phenylalanine were consumed completely after a cultivation time of 160 h. This results showed that the composition with respect to the amino acids might still be further optimized. The cell growth activity and glucose consumption could be modeled with semi-empirical balance equations based on Monod kinetics. The estimated parameters were μmax = 0.115 h^-1, Nmax =6.0×10^7 mL^-1. This study provided basic data for large scale production of heterologous proteins with Dictyostelium discoideum.[著者文摘]
